Temperature-sensitive mutants of bacteriophage mu.

چکیده
Temperature-sensitive mutants of bacteriophage Mu, which grow at 32 C but not at 42 C, have been isolated. These mutants fall into two groups. Group 1 mutants fail to lyse host cells at nonpermissive temperatures, whereas lysis occurs normally with the group 2 mutants. All of the group 1 mutants apparently belong to the cistrons mapping to the left of gene C, whereas the group 2 mutants have lesions in various genes between D and S.

C-terminal deletions can suppress temperature-sensitive mutations and change dominance in the phage Mu repressor.
Mutations in an N-terminal 70-amino acid domain of bacteriophage Mu's repressor cause temperature-sensitive DNA-binding activity. Surprisingly, amber mutations can conditionally correct the heat-sensitive defect in three mutant forms of the repressor gene, cts25 (D43-G), cts62 (R47-Q) and cts71 (M28-I), and in the appropriate bacterial host produce a heat-stable Sts phenotype (for survival of t...
متن کاملConstruction of the Recombinant Plasmid Expressing AID under the Control of Temperature-sensitive Promoter of Bacteriophage Lambda
Background and purpose: Activation-induced cytidine deaminase (AID) is a B-cell specific enzyme responsible for somatic hypermutation (SHM) and class switch recombination (CSR) of antibody genes within the B-cell follicle of peripheral lymphoid organs. Ectopic overexpression of the enzyme leads to mutations in non-B cells and Escherichia coli (E.coli) genes. However, induction of mutations in E...
متن کاملMutagenic effect of temperature-sensitive mutants of gene 42 (dCMP hydroxymethylase) of bacteriophage T4.
Certain temperature-sensitive mutants of gene 42 of bacteriophage T4 increase the reversion rates of some rII mutants in the same genome by about 4 to 10 times. This effect was usually found at 34 C, an intermediate permissive temperature, but not at 28 C.
متن کاملThe tRNA function of SsrA contributes to controlling repression of bacteriophage Mu prophage.
The small regulatory RNA SsrA has both tRNA and mRNA activities. It charges alanine and interacts with stalled ribosomes, allowing for translation to resume on the SsrA mRNA moiety. Hence, unfinished peptides carry a short amino acid tag, which serves as a signal for degradation by energy-dependent proteases. In SsrA-defective Escherichia coli strains, thermoinducible mutants of the transposabl...
متن کاملGenetic analysis of the developmental processes during germination and outgrowth of Bacillus subtilis spores with temperature-sensitive mutants.
HE developmental processes in bacteriophage T4 have been studied extenTsively with temperature-sensitive mutants and amber mutants (EDGAR, DENHARDT and EPSTEIN 1964; EDGAR and LIELAUSIS 1964). In Bacillus subtilis, the sporulation process has been studied with asporogenous mutants ( RYTER, SCHAEFFER and IONESCO 1966; TAKAHASHI 1965). We examined the developmental processes involved in the trans...
